موش تراریخت | Transgenic Mice

موش تراريخت چیست ؟!

تراريختگي یا ترانس ژنزیز (transgenesis) به فرآيند تغيير در ژنوم با استفاده از تکنیک های DNA نوترکيب گفته مي­شود که شامل موش تراریخت، موش ناک اوت و موش ناک این می باشد.

  • موش تراريخت (transgenic mice) به موشی گفته مي شود که ماده ژنتیکی جدید به ژنوم آن اضافه شده و صفت جديدي را کسب نموده است.
  • موش ناک اوت (knockout mice) به موشی گفته مي شود که ماده ژنتیکی از ژنوم آن حذف و یا دچار جهش شده است.
  • موش ناک این (knockin mice) به موشی گفته مي شود که ماده ژنتیکی از ژنوم آن حذف و ماده ژنتیکی دیگر جایگزین آن شده است.

مجموعه آزمایشگاه های سنجش و پایش کیاژن فارما با داشتن متخصصین خبره امکان تولید انواع مدل های حیوانی به طرق زیر را دارد:

  • توليد موش ترانس ژن از طريق ريزتزريقی به پيش هسته (pronucleus injection)

روش وازکتوم کردن موش چگونه است ؟!

در پروسه تولید موش تراریخت (transgenic mice)، از موش نر وازکتوم (vasectomized mice) جهت آماده سازی موش گیرنده جنین (foster/recipient) استفاده می شود.

  • حیوان را بیهوش کنید و در وضعیت پشتی قرار دهید.
  • پوست، عضلات و صفاق ناحیه خط میانی در مکان بالاتر از بخش آزاد آلت تناسلی را برش دهید.
  • یکی از بیضه ها را خارج کنید.
  • وازو دفران (vas deferens) را شناسایی و آن را ببرید یا بسوزانید.
  • بیضه را به داخل حفره شکمی برگردانید.
  • مراحل را بر روی بیضه دیگر تکرار کنید.
  • صفاق، عضلات و پوست را به صورت مجزا بخیه کنید.
