آزمون سمیت ژنی | Genotoxicity Test

آزمون سمیت ژنی (genotoxicity tests) با استفاده از تكنيك های كشت سلول، کشت میکروبی و مولکولی جهت تشخيص جهش هاي ژني، تغييرات در ساختمان و تعداد كروموزوم ها و ديگر اثرات سميیت بر DNA كه به علت تماس یک سیستم بیولوژیک با لوازم و تجهیزات پزشکی ممکن است حاصل ‏شود، بكار مي ‏رود.

در مجموعه آزمایشگاه های سنجش و پایش کیاژن فارما، تست سمیت ژنی (genotoxicity test) به 3 روش Comet assay ، Ames test و HPRT test بر اساس استانداردهای ISO 10993-3 ، OECD-471 و OECD-476 جهت ارزیابی موارد زیر بر روی لوازم و تجهیزات پزشکی  (medical devices)، دارو  (pharmaceuticals)، سم و آفت کش  (pesticides)، محصولات آرایشی و بهداشتی  (cosmetics)، مواد شیمیایی صنعتی (chemicals) و محصولات مصرفی (consumer products) انجام می شود.

  • تست کامت بوسیله الکتروفورز سلول های تحت تیمار با نمونه آزمایشی بر روی ژل آگاروز جهت بررسی تخریب DNA انجام می شود.
  • تست ایمز که به آزمون جهش زایی (mutagenicity test) نیز معروف است جهت بررسی پتانسیل موتاسیون نقطه ای (Point Mutations) ماده آزمایشی شامل جایگزینی (Substitution)، افزودن (Addition) یا حذف (Deletion) یک یا چند باز در ژن های سویه های باکتری سالمونلا تیفی موریوم (Salmonella Typhimurium) انجام می شود.
  • تست HPRT جهت ارزیابی جهش رو به جلو در و همچنین ارزیابی مواردی مانند جایگزینی جفت باز (base pair substitutions)، تغییر چارچوب خوانش (frameshifts) و تشخیص جهش‌ های ناشی از حذف ‌و درج کوچک (small deletions and insertions) بر روی لاین سلولی تخمدان همستر چینی (CHO) انجام می شود.
